RESUMEN

Semen proteins play an important role in male reproductive performance and sperm fertilization ability and can be used as potential biomarkers to evaluate male fertility. The role of cysteine-rich secretory protein 3 (CRISP3) in male reproduction remains unknown. This study aimed to investigate the role of CRISP3 in the reproductive performance of boars. Our results showed that the CRISP3 protein content was significantly and positively correlated with boar fertility, sow delivery rate, and litter size. CRISP3 is highly expressed in the bulbourethral gland of adult boars and is enriched in the seminal plasma. It is localized in the post-acrosomal region of the sperm head and migrates to the anterior end of the tail after capacitation. The CRISP3 recombinant protein did not affect sperm motility and cleavage rate, but it significantly downregulated the mRNA expression of inflammatory factors IL-α, IL-1ß, and IL-6 and the protein expression of IL-α and IL-6 in lipopolysaccharide (LPS)-induced RAW264.7 cells, indicating that CRISP3 has an immunomodulatory function. In conclusion, our study suggests that semen CRISP3 protein levels positively correlate with reproductive performance, which may be achieved by regulating immune responses in the female reproductive tract.

Neijiang (NJ) and Yacha (YC) are two indigenous pig breeds in the Sichuan basin of China, displaying higher resistance to diseases, lower lean ratio, and slower growth rate than the commercial Western pig breed Yorkshire (YS). The molecular mechanisms underlying the differences in growth and development between these pig breeds are still unknown. In the present study, five pigs from NJ, YC, and YS breeds were subjected to the whole genome resequencing, and then the differential single-nucleotide polymorphisms (SNPs) were screened using a 10-kb window sliding in 1-kb step using the Fst method. Finally, 48,924, 48,543, and 46,228 nonsynonymous single-nucleotide polymorphism loci (nsSNPs) were identified between NJ and YS, NJ and YC, and YC and YS, which highly or moderately affected 2,490, 800, and 444 genes, respectively. Moreover, three nsSNPs were detected in the genes of acetyl-CoA acetyltransferase 1 (ACAT1) insulin-like growth factor 2 receptor (IGF2R), insulin-like growth factor 2 and mRNA-binding protein 3 (IGF2BP3), which potentially affected the transformation of acetyl-CoA to acetoacetyl-CoA and the normal functions of the insulin signaling pathways. Moreover, serous determinations revealed significantly lower acetyl-CoA content in YC than in YS, supporting that ACAT1 might be a reason explaining the differences in growth and development between YC and YS breeds. Contents of phosphatidylcholine (PC) and phosphatidic acid (PA) significantly differed between the pig breeds, suggesting that glycerophospholipid metabolism might be another reason for the differences between Chinese and Western pig breeds. Overall, these results might contribute basic information to understand the genetic differences determining the phenotypical traits in pigs.

The objectives were to evaluate the effects of standardized ileal digestible (SID) His:Lys ratio above the current NRC requirement on growth performance, intestinal health, and mobilization of His-containing proteins, including hemoglobin, carnosine, and trypsinogen, in nursery pigs from 7 to 11 kg body weight (BW). Forty pigs (26 d of age; initial BW of 7.1 ±â€…0.5 kg) were allotted to 5 dietary treatments based on a randomized complete block design with sex and initial BW as blocks. Dietary treatments were supplemented with varying SID His to Lys ratios of 26%, 32%, 38%, 43%, and 49% and fed to pigs for 14 d (SID Lys = 1.22%). Feed intake and BW were recorded at d 0, 7, and 14 to measure growth performance. Blood samples were collected on d 12. Pigs were euthanized on d 14 to collect pancreas, longissimus dorsi muscles, mid-jejunum, and jejunal mucosa. Data were analyzed using the Proc Mixed of SAS. Growth performance was not affected, whereas varying SID His to Lys ratio affected hemoglobin (P < 0.05, max: 12 g/dL at 36%), immunoglobulin A (IgA, P < 0.05, min: 1.25 µg/mg at 35%) in jejunal mucosa, villus height (P = 0.065, max: 536 µm at 40%) in jejunum, trypsinogen (P = 0.083, max: 242 pg/mg at 41%) in pancreas, and carnosine (P = 0.051, max: 4.7 ng/mg at 38%) in muscles. Varying SID His to Lys ratios linearly increased (P < 0.05, from 1.95 to 2.80 nmol/mg) protein carbonyl in muscles and decreased (P < 0.05, from 29.1% to 26.9%) enterocyte proliferation. In conclusion, SID His to Lys ratio between 35% and 41% in diets fed to nursery pigs at 7 to 11 kg enhanced intestinal health and maximized concentrations of His-containing proteins, indicating that His-containing proteins are effective response criteria when determining His requirement.


Histidine is an essential amino acid for protein synthesis, but it also plays a vital role in the metabolic system of pigs. An accurate assessment of His requirement provides pivotal information for efficient growth and health of pigs. Growth performance and plasma His concentration have been used to assess His requirement, but they may not be the effective parameters due to the contribution of His from mobilization of His-containing proteins, such as hemoglobin, carnosine, and pancreatic enzymes. Hemoglobin is a transport protein and the main component in red blood cells, enabling oxygen transport throughout the body. Most carnosine is stored in muscles at 3 to 4 g/kg wet weight and has antioxidative effects to prevent cells from oxidative damages. In addition, His has a critical role in serine peptidases as a part of the catalytic triad. In this study, growth performance did not respond to His deficiency due to the compensation of His from His-containing proteins and potentially due to a short experimental period. Standardized ileal digestible His to Lys ratio between 35% and 41% maximized concentrations of His-containing proteins and enhanced intestinal health in pigs at 7 to 11 kg body weight. This study indicated that hemoglobin, carnosine, and trypsinogen are effective response criteria when determining His requirement.

Sujiang pigs are a synthetic breed derived from Jiangquhai, Fengjing, and Duroc pigs. In this study, we sequenced the genome of 62 pigs with a coverage depth of 10× to 20×, including 27 Sujiang and 35 founder breed pigs, and we collected 360 global pigs' genome sequence data from public databases including 39 Duroc pigs. We obtained a high-quality variant dataset of 365 Sujiang pigs by imputing the porcine 80 K single nucleotide polymorphism (SNP) Beadchip to the whole-genome scale with a total of 422 pigs as a reference panel. A dataset of 365 imputated Sujiang pigs was used to perform single-trait genome-wide association study (GWAS) and meta-analyses for growth and fatness traits. Single-trait GWAS identified 1 907, 18, and 14 SNPs surpassing the suggestively significant threshold for backfat thickness, chest circumference, and chest width, respectively. Meta-analyses identified 2 400 genome-wide significant SNPs and 520 suggestively significant SNPs for backfat thickness and chest circumference, and 719 genome-wide significant SNPs and 1 225 suggestively significant SNPs for all seven traits. According to the meta-analysis of backfat thickness and chest circumference, a remarkable region of 2.69 Mb on Sus scrofa chromosome 4 containing FAM110B, IMPAD1, LYN, MOS, PENK, PLAG1, SDR16C5 and XKR4 was identified as a candidate region. The haplotype heat map of the 2.69 Mb region verified that Sujiang pigs were derived from Duroc and Chinese indigenous pigs, especially Jiangquhai pigs. The Kruskal-Wallis test showed that haplotypes of the 2.69 Mb region significantly affected backfat thickness and chest circumference traits. We then focused on PLAG1, an important growth-related gene, and identified two synonymous SNPs with obvious differences among different breeds in the PLAG1 gene. We then performed genotyping of 365 Sujiang, 150 Duroc, 95 Jiangquhai, and 100 Fengjing pigs to confirm the above result and verified that the two variants significantly affected phenotypes of growth and fatness traits. Our findings not only provide insights into the genetic architecture of porcine growth and fatness traits but also provide potential markers for selective breeding of these traits in Sujiang pigs.

Improving productivity is an urgent issue in the swine industry if it is to compete internationally. However, lack of data about recent productivity transition obstructs stakeholder planning. This study investigated the yearly productivity trends among farrow-to-finish swine farms in Japan using annual productivity data from 2013-2018 obtained for 70 farms in Japan. The productivity parameters analyzed were pigs born alive per litter (PBA), preweaning mortality (PRWM), pigs weaned per litter (PWL), litters per mated female per year (LMFY), pigs weaned per mated female per year (PWMFY), post-weaning mortality (POWM) and marketed pigs per mated female per year (MP). Data were classified into three groups based on the size of the average female inventory and compared among groups. Results presented the mean PBA increased continuously over the 6-year period (P<0.001), and the PWL, PWMFY, and MP means began increasing after 2015 (P<0.001). These upward trends were particularly remarkable on large farms. The mean PRWM increased sharply in 2014, thus inhibiting the increases in PWL, PWMFY, and MP for the same year. The LMFY and POWM means did not change during the study period. Altogether, productivity in Japan improved markedly during the study period, indicating highly prolific sows were well utilized with suitable breeding techniques among farmers these days. Continued genetic improvement and sow management would aid further development in Japan.

The microbiota hosted in the pig gastrointestinal tract are important to health of this biomedical model. However, the individual species and functional repertoires that make up the pig gut microbiome remain largely undefined. Here we comprehensively investigated the genomes and functions of the piglet gut microbiome using culture-based and metagenomics approaches. A collection included 266 cultured genomes and 482 metagenome-assembled genomes (MAGs) that were clustered to 428 species across 10 phyla was established. Among these clustered species, 333 genomes represent potential new species. Less matches between cultured genomes and MAGs revealed a substantial bias for the acquisition of reference genomes by the two strategies. Glycoside hydrolases was the dominant category of carbohydrate-active enzymes. Four-hundred forty-five secondary metabolite biosynthetic genes were predicted from 292 genomes with bacteriocin being the most. Pan genome analysis of Limosilactobacillus reuteri uncover the biosynthesis of reuterin was strain-specific and the production was experimentally determined. This study provides a comprehensive view of the microbiome composition and the function landscape of the gut of weanling piglets and a valuable bacterial resource for further experimentations. IMPORTANCE The microorganism communities resided in mammalian gastrointestinal tract impacted the health and disease of the host. Our study complements metagenomic analysis with culture-based approach to establish a bacteria and genome collection and comprehensively investigate the microbiome composition and function of the gut of weanling piglets. We provide a valuable resource for further study of gut microbiota of weanling piglet and development of probiotics for prevention of disease.

Social behavior is a key component of pig welfare on farms, but little is known on the development of social behaviors in piglets. This study aimed to explore social behaviors and identify early social styles in suckling piglets. Social behaviors of 68 piglets from 12 litters were scored continuously for 8 h per day at 21 and 42 days of age, and were included in a Hierarchical Clustering on Principal Components analysis to identify clusters of pigs with similar social styles. Social nosing represented 78% of all social interactions given. Three social styles were identified: low-solicited inactive animals (inactive), active animals (active), and highly-solicited avoiders (avoiders). Belonging to a cluster was independent of age, but was influenced by sex, with females being more represented in the 'inactive' cluster, and males in the 'active' cluster, whereas both sexes were equally represented in the 'avoider' cluster. Stability of piglets' allocation to specific clusters over age was high in the 'inactive' (59%) and 'active' (65%) clusters, but low in the 'avoider' cluster (7%). Haptoglobin and growth rate were higher in 'active' than 'inactive' pigs, and intermediate in 'avoiders'. Our findings suggest the existence of transient social styles in piglets, likely reflective of sexual dimorphism or health status.

Insects represent a promising alternative source to more expensive components of the feeding regimen of farm animals for meat production. With this aim in mind, we conducted this experiment to investigate the effects of dried larvae of darkling beetles (Zophobas morio L.) and larvae of wax moths (Galleria mellonella L.) on growth performance, nutrient digestibility and blood profiles in growing piglets. A total of 27 crossbred piglets ((LWхL)хD), 39 days of age and 14.39±0.19 kg of body weight) were randomly divided into three groups (n=9 per group) based on gender and body weight. We substituted part of a fishmeal and a grain with 2.5% of dried larvae of darkling beetles (Zophobas morio L.) or with 3.0% of dried larvae of wax moths (Galleria mellonella L.). The replacement of fishmeal with insect biomass in the feeding regimen of the pigs did not change growth rate among groups and did not altered the digestibility of dry matter, crude protein and crude fiber (p ˃ 0.05). At the same time, the digestibility of the fat has increased in the experimental groups with dry larvae (p ≤ 0.05). Blood counts in all groups were within the physiological norm. As compared to control group, piglets of the experimental groups had an increase in bactericidal, lysozyme activity of the blood serum and phagocytic activity of neutrophils. Thus, supplementation of dry larvae of superworm or waxworm in the ration didn't affect negatively growth performance of experimental piglets while improving their parameters of non-specific immunity.

BACKGROUND: The interplay between the gut microbiota and feeding behavior has consequences for host metabolism and health. The present study aimed to explore gut microbiota overall influence on feeding behavior traits and to identify specific microbes associated with the traits in three commercial swine breeds at three growth stages. Feeding behavior measures were obtained from 651 pigs of three breeds (Duroc, Landrace, and Large White) from an average 73 to 163 days of age. Seven feeding behavior traits covered the information of feed intake, feeder occupation time, feeding rate, and the number of visits to the feeder. Rectal swabs were collected from each pig at 73 ± 3, 123 ± 4, and 158 ± 4 days of age. DNA was extracted and subjected to 16 S rRNA gene sequencing. RESULTS: Differences in feeding behavior traits among breeds during each period were found. The proportion of phenotypic variances of feeding behavior explained by the gut microbial composition was small to moderate (ranged from 0.09 to 0.31). A total of 21, 10, and 35 amplicon sequence variants were found to be significantly (q-value < 0.05) associated with feeding behavior traits for Duroc, Landrace, and Large White across the three sampling time points. The identified amplicon sequence variants were annotated to five phyla, with Firmicutes being the most abundant. Those amplicon sequence variants were assigned to 28 genera, mainly including Christensenellaceae_R-7_group, Ruminococcaceae_UCG-004, Dorea, Ruminococcaceae_UCG-014, and Marvinbryantia. CONCLUSIONS: This study demonstrated the importance of the gut microbial composition in interacting with the host feeding behavior and identified multiple archaea and bacteria associated with feeding behavior measures in pigs from either Duroc, Landrace, or Large White breeds at three growth stages. Our study provides insight into the interaction between gut microbiota and feeding behavior and highlights the genetic background and age effects in swine microbial studies.

Apart from improving husbandry conditions and animal welfare, there is a clear public demand to increase transparency in agricultural activities. Personal farm tours have shown to be appreciated by citizens but are limited in their impact because of hygiene requirements and accessibility. Virtual farm tours are a promising approach to overcome these limitations but evidence on their perceptions is missing. This study analyzes how a virtual farm tour is perceived by showing participants (n = 17) a 360-degree video of a conventional pig fattening pen on a tablet and via virtual reality (VR) glasses. Semi-structured in-depth interviews were conducted to analyze perceptions and level of immersion and to elicit differences between media devices. Participants' perception of the pig fattening pen was rather poor and depended on the recording perspective as well as on the media device. However, housing conditions were perceived more positively compared to the image participants had in mind prior to the study, and thus the stable was considered as a rather positive example. Participants described virtual farm tours as suitable tool to improve transparency and information transfer and to gain insights into husbandry conditions. They appreciated the comfortable and entertaining character of both media devices and named various possibilities for implementation. However, VR glasses were favored regarding the higher realistic and entertaining value, while the tablet was considered beneficial in terms of usability. The presentation of video sequences without additional explanations about the farm or the housing conditions were claimed insufficient to give an adequate understanding of the seen content.

Heat stress affects pig metabolism, health and welfare, resulting in reduced growth and important economic losses. The present experiment aimed to evaluate the effects of two climatic environments [temperate (TEMP) vs. tropical humid (TROP)] on feeding behaviour in growing pigs. The feeding behaviour traits were measured with automated feeders and included: daily feed intake, daily eating time, feeding rate, daily number of meals, feed intake per meal, and feeding time per meal. Pigs came from a backcross population between Large White (LW, heat sensitive) and Creole (CR, heat tolerant) pigs. The same 10 F1 LW × CR boars (sire families [SF]) were mated with related LW sows in each environment. Feeding behaviour was recorded for a total of 1,296 pigs (n = 634 pigs for TEMP and n = 662 pigs for TROP) between 11 and 23 weeks of age. Growth performance and thermoregulatory responses (rectal and skin temperatures) were also measured. Results show that TROP conditions affect feeding behaviour traits: animals had more meals per day but these meals were smaller both in duration and in size, resulting in lower daily feed intake and less time eating per day. Significant SF by environment (GxE) interactions were found for all feeding behaviour traits. When SF were distributed into robust and sensitive groups (previously defined according to performance and thermoregulatory traits), results showed group by environment interactions for all feeding traits, except meal frequency. Moreover, a significant difference in feeding rate between robust and sensitive group was detected in TEMP, suggesting that feeding rate may be a good candidate to evaluate heat tolerance.

The type of myofiber is related to the quality of meat. The slow oxidized myofiber helps to increase the tenderness and juiciness of muscle. Numerous studies have shown that circRNA plays a key role in skeletal muscle development. However, the role of circRNA in porcine skeletal myofiber types is unclear. In this study, we performed high-throughput RNA sequencing to study the differential expression of circRNA in the longissimus dorsi and the soleus muscle. A total of 40,757 circRNAs were identified, of which 181 were significantly different. Interestingly, some circRNAs were involved in metabolism pathways, AMPK, FoxO, and PI3K-Akt signaling pathways. Besides, we focused on a novel circRNA-circMYLK4. By injecting circMYLK4-AAV into piglets, we found that circMYLK4 significantly increased the mRNA and protein levels of the slow muscle marker genes. In summary, our study laid an essential foundation for further research of circRNA in myofiber type conversion and higher meat quality.

Taking into account that in the literature on pork-production Life Cycle Assessment (LCA) there are a few studies about the Iberian pig, the present article evaluates an extensive (growing-fattening) Iberian-pig system in Spain, producing meat for Iberian ham and other quality-labelled products. The study has been based on Cumulative Energy Demand (CED), Global Warming Potential (GWP), ReCiPe (midpoint; endpoint) and USEtox (human toxicity; ecotoxicity). The analysis involves feed (for pigs and piglets), transportation, drinking water, straw usage and building materials (concrete). The impacts have been evaluated per kg of live or carcass weight (two functional units). The results show that the total impacts (per kg of live or carcass weight) range from: 1) 22.05 to 28.19 MJprim (CED), 2) 4.37 to 6.19 kg CO2.eq (GWP 20a, 100a and 500a), 3) 0.86 to 1.08 Pts (ReCiPe endpoint single-score, involving Human health, Ecosystems and Resources), 4) 9.9 × 10-6 to 1.2 × 10-5 DALY (ReCiPe endpoint with characterisation), 5) 2.8 × 10-7 to 3.5 × 10-7 (species.yr) (ReCiPe endpoint with characterisation), 6) 10.12 to 12.66 CTUe (USEtox: ecotoxicity). Overall, the results show that the feed for the pigs is responsible for the major part of the environmental impacts. More analytically, maize and soya are the components with the highest environmental impacts due to factors such as transportation, use of fertilisers and diesel fuel. The discussion about pig-production environmental impacts and the role of extensive pig farming is enriched with comparisons with the literature on pig-production LCA. Critical parameters are identified and discussed, with the aim of proposing solutions to reduce pork-production environmental impacts. Finally, the usefulness of the present study and future prospects are presented.

The present study was conducted to evaluate the effect of cassava hard pellet and residue pellet-based diet as a replacement for corn on the growth performance, total tract digestibility and blood profile in pigs. Three experiments were conducted to evaluate the processed cassava as replacement for corn in pigs. In Exp. 1, seventy-two weanling pigs with an average initial body weight of 6.44 ± 0.52 kg were assigned to one of the two treatments: CON, basal diet, and CHP, basal diet with cassava hard pellet for 28 days trial. The dietary inclusion of CHP had significantly decreased average daily gain of weanling pigs during d 1-14 and the overall experimental period. In addition, significant reduction was observed on gain-to-feed ratio during d 1-14, while no effect was noted on average daily feed intake. Exp. 2 was carried out for 42 days, in which 84 growing pigs were randomly assigned to 1 of 3 treatments with 4 pigs/pen and 7 pen/treatment. The three dietary treatments were supplement with 0%, 20% and 40% of cassava. Dietary inclusion of cassava supplementation has failed to affect the growth performance and blood profile during overall experiment. In Exp. 3, a total of 96 crossbred finishing pigs were randomly assigned to one of four dietary treatments containing 25% cassava in a 2 × 2 factorial design with two different levels of energy with or without 0.1% carbohydrase for 28 days. Pigs fed 0.1% carbohydrase with high energy diet reduced average daily feed intake and increased gain-to-feed ratio compared to those fed low energy diet. However, significantly enhanced nutrient digestibility in both energy and carbohydrase groups were observed. In summary, the inclusion of cassava basal diet with 0.1% carbohydrase supplementation has beneficially enhanced the growth performance and nutrient digestibility of pigs.

Testicular aromatase catalyzes the synthesis of estradiol, which contributes to regulation of porcine Sertoli cell proliferation and postpubertal maintenance of Sertoli cell numbers. Although aromatase enzymatic activity decreases with age and is persistently reprogrammed by prepubertal treatment with the aromatase inhibitor letrozole, the molecular bases for regulation have not been identified. DNA methylation was examined as a potential regulatory mechanism using DNA from Leydig cells isolated from 16-, 40-, and 68-week-old boars and from 68- week-old littermates treated with the aromatase inhibitor, letrozole. Methylation levels of individual CpG dinucleotides located in the distal untranslated exon 1 of the relevant aromatase encoding gene, CYP19A3, were quite high in Leydig cell DNA, and increased further with maturity of boar (P < 0.05), while aromatase activity and transcript abundance decreased more than two-fold. However, reduced aromatase activity following letrozole treatment was not accompanied by altered DNA methylation. Testicular expression of miR378 was altered by prepubertal treatment with letrozole. The data provide evidence for two different epigenetic mechanisms that regulate aromatase expression and enzymatic activity in the boar testis.

BACKGROUND: The beneficial function of phytase and 25-hydroxyvitamin D3 (HyD) on the feed utilization rate has been widely investigated. However, studies concerning its influence on weaned piglets largely lag behind. The aim of this study was to investigate the effects of phytase and HyD supplementation on the growth performance and bone development in weaned piglets under dietary Ca and P deficiency. RESULTS: The results showed that dietary Ca and P deficiency decreased (P < 0.05) the content of serum P in 6-10 kg piglets, as well as reducing (P < 0.05) the contents of serum Ca and P, average daily gain (ADG), bone mineral density (BMD), breaking force (BF), bone ash and femur Ca in 10-20 kg piglets. Compared with the control group, the feed-to-gain ratio (F/G) of 6-10 kg piglets in the Phy group was decreased (P < 0.05), whereas the ADG, blood Ca and P, BMD, BF, bone ash, P apparent digestibility, Ca and P retention rate of 10-20 kg piglets were increased (P < 0.05). The contents of serum osteocalcin and HyD in 6-10 kg piglets and ADG were higher than in the control group (P < 0.05), as well as the contents of serum Ca and HyD in 10-20 kg piglets in the HyD treatment group. Supplementation with both Phy and HyD decreased the F/D (P < 0.05) and increased the contents of serum Ca, P and HyD in 6-10 kg piglets as well as enhancing the ADG, BMD, BF, bone ash, femur Ca and P, serum Ca and P, HyD, and the apparent digestibility and retention of Ca and P (P < 0.05) in 10-20 kg piglets. Supplementation with Phy and HyD in Ca- and P-deficient dietary decreased bone resorption, and improved tight arrangement of collagen fibers and oblique fibers in weaned piglets. CONCLUSION: These data indicated that supplementation with both 1500 U kg-1 Phy and 50 µg kg-1 HyD could enhance dietary Ca and P utilization and promote bone development in low Ca and P dietary, and supplementation with both Phy and HyD had a significant synergy effect compared to single supplement. © 2021 Society of Chemical Industry.

BACKGROUND: With the development of intensive farming, long-term exposure of pigs to poor light conditions is not conducive to the production of vitamin D3 , and vitamin D3 deficiency could affect absorption and metabolism of calcium (Ca) and phosphorus (P). 25-Hydroxyvitamin D3 (25OHD3 ) has higher bioactivity than regular vitamin D3 . This study investigated the effects of 25OHD3 on performance, serum parameters, fecal microbiota, and metabolites in weaned piglets fed with low Ca-P diet. RESULTS: It was found that a low Ca-P diet supplemented with 50 µg/kg 25OHD3 (NC + 25-D) improved (P < 0.05) average daily gain (ADG) in phase 2 and in the overall period of the experiment, and increased (P < 0.05) the immunoglobulin G (IgG), immunoglobulin A (IgA), catalase (CAT), bone-specific alkaline phosphatase (BALP), and osteocalcin (OC) serum content on day 28 compared with a low Ca-P diet (NC), but no differences were observed between a normal Ca-P diet (PC) and the NC + 25-D diet. Compared with NC, the abundance of Firmicutes was higher (P < 0.05) in PC and NC + 25-D. NC + 25-D decreased (P < 0.05) the abundance of Streptococcaceae compared with PC and NC, and increased (P < 0.05) the abundance of Lachnospiraceae compared with NC. Serum 25OHD3 was negatively correlated with the abundance of fecal Streptococcaceae (P < 0.05), and positively correlated with the abundance of fecal Lachnospiraceae (P < 0.05). CONCLUSION: Supplementation of 25OHD3 in a low Ca-P diet improved serum immunity, bone biochemical parameters, and fecal microbiota such as decreased Streptococcaceae abundance and increased Lachnospiraceae abundance, which could subsequently promote growth of piglets. The effects were similar to that of a normal Ca-P diet. © 2021 Society of Chemical Industry.

Fumonisin B1 (FB1) is the most common food-borne mycotoxin produced by the Fusarium species, posing a potential threat to human and animal health. Pigs are more sensitive to FB1 ingested from feed compared to other farmed livestock. Enzymatic degradation is an ideal detoxification method that has attracted much attention. This study aimed to explore the functional characteristics of the carboxylesterase FumDSB in growing pigs from the perspective of brain-gut regulation. A total of 24 growing pigs were divided into three groups. The control group was fed a basal diet, the FB1 group was supplemented with FB1 at 5 mg/kg feed, and the FumDSB group received added FumDSB based on the diet of the FB1 group. After 35 days of animal trials, samples from the hypothalamus and jejunum were analyzed through HE staining, qRT-PCR and immunohistochemistry. The results demonstrated that the ingestion of FB1 can reduce the feed intake and weight gain of growing pigs, indicating that several appetite-related brain-gut peptides (including NPY, PYY, ghrelin and obestatin, etc.) play important roles in the anorexia response induced by FB1. After adding FumDSB as detoxifying enzymes, however, the anorexia effects of FB1 were alleviated, and the expression and distribution of the corresponding brain-gut peptides exhibited a certain degree of regulation. In conclusion, the addition of FumDSB can reduce the anorexia effects of FB1 by regulating several brain-gut peptides in both the hypothalamus and the jejunum of growing pigs.

This study aimed to evaluate the effects of a sequential feeding program (SEQ) with diets varying in amino acid (AA) concentrations in the first and last 12 h of the day on the performance, body composition, and nutrient balance of growing-finishing pigs. Sixty-eight castrated male pigs were distributed in four treatments: a daily feeding program (DP) and three SEQs. In the DP, dietary requirements of AA were adjusted daily. In the SEQ, dietary daily requirements of AA were adjusted every 12 h, providing a low AA concentration in period 1 (P1; 00:00-11:59 h) and a high AA concentration in period 2 (P2; 12:00-23:59 h). In the SEQ, three different levels of low and high AA concentrations were evaluated: ±20%, ±30%, and ±40%. The experiment lasted 82 days and was divided into phase 1 (25-50 kg body weight; BW), phase 2 (50-70 kg BW), and phase 3 (70-100 kg BW). During phase 1, irrespective of dietary AA concentration, SEQ pigs had higher lysine intake, protein gain, and phosphorus efficiency than DP pigs (P ≤ 0.05). Pigs in the SEQ showed a tendency for greater average daily gain, body protein, and body lipids compared to the DP pigs (P ≤ 0.10). During phase 2, SEQ pigs showed a tendency for higher average feed intake in P2 compared to DP pigs (P = 0.07); consequently, average daily gain, body protein, and phosphorus retention tended to increase (P ≤ 0.10). During phase 3, SEQ pigs had a higher average feed intake in P2 than DP pigs (P = 0.03). However, they had a similar body composition (P > 0.05) and a tendency for higher nitrogen excretion (P = 0.06) than DP pigs. Our results suggest that SEQ is an effective approach for improving the performance and body composition of growing pigs.

The hog industry is the core industry in the field of agriculture and animal husbandry in China, which development is related to the improvement of people's quality of life. The production of the hog industry has been restricted by environmental regulations, which needs to reduce costs and improve efficiency. Based on the data of 29 provinces from 2008 to 2019, this paper aims to use the stochastic frontier analysis method to calculate the cost efficiency of fattening pigs, sows, and piglets in three stages of pig production and focuses on the impact of environmental regulation policies on cost efficiency. The study found that the cost efficiency of fattening pigs, sows, and piglets in China were 0.77, 0.79, and 0.53, respectively, and the efficiency losses were 23%, 21%, and 47%, respectively. The impact of environmental regulation policies on the cost efficiency of fattening pigs showed an ' N ' trend, and the impact on the cost efficiency of sows and piglets showed an inverted ' N ' trend. For fattening pigs, increasing the intensity of environmental regulation, and exceeding the second inflection point of the ' N ' curve can achieve the dual goals of environmental governance and cost reduction and efficiency increase. For sows, reducing the intensity of environmental regulation appropriately can avoid cost-efficiency loss. For piglets, environmental regulation policies have not effectively incentives the cost efficiency of piglets. In addition, raising the level of technology investment in fattening pigs and sows can achieve cost efficiency gains, and can farmers use emerging financial product tools such as ' insurance + futures ' to avoid market risks and efficiency losses.
